ABSTRACT

Objective: To investigate the chemical constituents of n-butanol fraction of the ethanol extracts from spray of Gentianae szechenyii. Methods: The chemical constituents were isolated and purified by repeated preparative high-performance liquid chromatography. The structures of the isolated compounds were identified on the basis of extensive spectroscopic techniques. Results: Six compounds were obtained in n-butanol fraction of the ethanol extracts from G. szechenyii and identified as gentizechenlioside A (1), szechenyin A (2), isoscoparin-2″-O-glucopyranoside (3), isoscoparin (4), gentiournosides D (5), and depressine (6). Conclusion: Compound 1 is a new iridoid glycoside which named as gentizechenlioside A.

ABSTRACT

OBJECTIVE:To establish a method for the simultaneous determination of loganic acid and isoscoparin in Sanwei longdanhua tablets. METHODS:HPLC method was adopted. The determination was performed on Inertsil ODS-3 column with mobile phase consisted of methanol-0.2% phosphoric acid (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was set at 240 nm,and column temperature was 30 ℃. The sample size was 10 μL. RESULTS:The linear range were 0.040 08-4.008 0 μg(r=0.999 9)for loganic acid and 0.021 96-2.196 0 μg(r=0.999 9)for isoscoparin. The quantitative limits were 0.160 32 and 0.087 8 ng/mL,and detection limits were 0.080 16 and 0.043 92 ng/mL. RSDs of precision,stability and reproducibility tests were all lower than 2%. The recoveries were 103.07%-104.26%(RSD=0.52%,n=6) and 95.57%-99.61%(RSD=1.55%,n=6). CONCLUSIONS:The method is simple,accurate and suitable for simultaneous determination of loganic acid and isoscoparin in Sanwei longdanhua tablets.

ABSTRACT

Objective: To study the chemical constituents from the whole plant of Swertia davidi. Methods: The chemical constituents were isolated and purified with silica gel column chromatography, silica gel chromatography, etc. The structures of the compounds were identified by physicochemical properties and various spectroscopic methods. Results: Sixteen compounds were isolated as uvaol (1), oleanolic acid (2), 5-carboxyl-3, 4-dihydrogen-1H - 2-benzopyran-1-one (3), 3β, 12β-pregnane-16-12-ene-20 (4), swertisin (5), 6-C-β-glucopyranosyl-7-O-methylluteolinidin (6), isoscoparin (7), 6-demethoxy-7-methylcapillarisin (8), 3-acetoxy-28-hydro-xyl-12-eneurs (9), gentiopicroside (10), N-pentacosy-2-carboxy-benzoylamide (11), 4-O-glycosyloxy-2-hydroxy-6-methoxy-acetophenone (12), quercetin-3-O-β-D-xylopyranosyl-(1→2) - β-D-galactopyranoside (13), 8-epikingiside (14), 3, 5-dicaffeoylquinicacid (15), and 2-methylphenyl-1-O-β-D-glucopyranoside (16). Conclusion: All the compounds are isolated from S. davidi for the first time.
